CB10 1SD, Hinxton, UK)Multiple sclerosis (MS) is a disease of the central nervous system that is a common cause of chronic neurological disability. The activation of autoreactive T cells leads to inflammation and recognition of antigens such as Myelin Basic Protein, a component of the axons protective sheath. Several genetic risk variants, environmental insults and signalling pathways have been proposed, but to date the full mechanism that mediates this activation is unknown. CombiMS is a consortium of experimental, clinic and mathematical modeling groups that aims to understand how signaling is deregulated in MS. We have characterized the MS-immune signaling network using publicly available data and newly established phosphoproteomic and genotypic assays. Next, we seek to further develop a mathematical framework previously established by us, using a data-driven logic formalism predictive of network topology. To that end, we hope the VizBi 2014 will be the perfect opportunity to discuss how to link these (and others?) layers of data to improve visualization of MS-immune signaling mechanisms, thereby contributing to elucidate the activation mechanisms of MS.
